When I try to playtest my game, the Spatial Voice button at the top-left corner of my screen along with “Connect” and Chat doesn’t appear, this wasn’t an issue before the new updated ROBLOX menu.
This happens in both Solo playtesting and in a local server.
Some notes I just want to clarify:
The game is published to ROBLOX, and has microphone communication turned on.

thanks for reporting this issue. The fix for the issue triggering that error output you noted should go live with the next studio release (current ETA Thursday).
(The issue happens in Studio on Windows, not on Mac, so if you have a Mac machine, you could switch to that in the meantime)
That issue being fixed soon for Windows Studio aside, currently in general Voice Chat testing in Studio is only supported in Team Create Team Test, not in Local Server Play Solo/Play MP.

just wanted to give a heads-up: the latest Studio update is up and with that you should not be getting those errors on Windows anymore and should be able to test voice chat in Team Create Team Test.
